Check whether a bus goes to a destination and ask which numbered bus to take.

The idea

に marks the destination with 行きます: 駅に行きますか asks whether it goes to the station. どの comes before a noun: どのバス asks which bus out of the available choices. 三番(さんばん) identifies number three; 三番のバス refers to bus number three.
